5株亚硝化反硝化菌的筛选及缺氧吸磷特性研究_金鹏

书5株亚硝化反硝化菌的筛选及缺氧吸磷特性研究金鹏(中国建筑东北设计研究院有限公司,辽宁沈阳110006)摘要:在已稳定运行的连续流双污泥亚硝化反硝化除磷系统的缺氧池中取污泥,经亚硝化反硝化菌选择培养基培养后,筛选出5株亚硝化反硝化菌:FA5、FA6、FB2、FB4和FB5,对其进行革兰氏染色和一系列生理生化试验,确定其分别为克雷伯氏菌属、芽孢杆菌属、肠杆菌属、莫拉氏菌属和芽孢杆菌属。考察4种亚硝化反硝化菌的厌氧释磷和缺氧吸磷特性,同时进行PHB染色和异染颗粒染色。结果表明,4种亚硝化反硝化菌中都含有聚磷和PHB颗粒,且都有厌氧释磷和缺氧吸磷现象,即4种菌都能进行同步反硝化除磷,但芽孢杆菌属、克雷伯氏菌属、莫拉氏菌属和肠杆菌属的吸磷量依次减少,吸磷量分别为(1.48×10-11)、(1.13×10-11)、(6.28×10-12)和(6.05×10-12)mg/cfu。关键词:双污泥系统;亚硝化反硝化除磷;亚硝化反硝化菌;缺氧吸磷中图分类号:X703文献标识码:A文章编号:1000-4602(2014)05-0033-04基金项目:中建股份科技研发课题(CSCEC-2012-Z-14)ScreeningofFiveNitrosationDenitrifyingOrganismsandCharacteristicsofAnoxicPhosphorusUptakeJINPeng(ChinaNortheastArchitecturalDesignandResearchInstituteCo.Ltd.,Shenyang110006,China)Abstract:Sludgewastakenfromtheanoxicreactorofacontinuousflowdoublesludgenitrosationdenitrifyingphosphorusremovalsystemwhichhadbeenoperatingstably.Nitrosationdenitrifyingorgan-ismswerescreenedbytheselectivemediumofnitrosationdenitrifyingorganisms.Fivestrainsofnitrosati-ondenitrifyingorganisms(FA5,FA6,FB2,FB4andFB5)wereidentifiedbyGram’sstainingandase-riesofphysiologicalandbiochemicalexperimentstobeKlebsiella,Bacillus,Enterobacter,MoraxellaandBacillusrespectively.Thecharacteristicsofanaerobicphosphorusreleaseandanoxicphosphorusuptake,whendyeingwithpoly-β-hydroxybutyricacid(PHB)andpoly-phosphate,forfourkindsofdenitrifyingorganismswerestudiedsimultaneously.TheresultsshowedthattherewerebothPHBandpoly-phosphateinfournitrosationdenitrifyingorganismsandallhadmanifestedanaerobicphosphorusreleaseandanoxicphosphorusuptake.Thatis,thefourorganismscoulddenitrifyandremovephosphorussimultaneously.However,thequantitiesofphosphorusuptakeofBacillus,Klebsiella,MoraxellaandEnterobacterwere1.48×10-11mg/cfu,1.13×10-11mg/cfu,6.28×10-12mg/cfuand6.05×10-12mg/cfu,respective-ly,showingagradualdecrease.Keywords:doublesludgesystem;nitrosationdenitrifyingphosphorusremoval;nitrosationde-nitrifyingorganisms;anoxicphosphorusuptake·33·第30卷第5期2014年3月中国给水排水CHINAWATER&WASTEWATERVol.30No.5Mar.2014亚硝化反硝化除磷(NDPR)是指在厌氧/缺氧交替运行条件下能够在缺氧阶段以亚硝态氮为电
