桑葚多糖的分离纯化及组成分析

华西药学杂志WCJ·PS2014,29(4)∶401~404作者简介:田仁君(1978—),女,硕士,讲师,从事天然药物活性成分的分离及研究工作。Email:tianrenjun888@163.com桑葚多糖的分离纯化及组成分析田仁君(四川中医药高等专科学校,四川绵阳621000)摘要:目的探讨桑葚多糖(PMF)的分离纯化方法,研究纯化组分的组成及结构。方法首先采用DEAE-52纤维素离子交换柱进行分级洗脱纯化PMF,再用SephadexG-100葡聚糖凝胶色谱柱进一步纯化得纯化多糖;用苯酚-硫酸法测定总糖的含量、考马斯亮蓝法测定蛋白质的含量、硫酸-间羟联苯法测定糖醛酸的含量、氯化钡-明胶比色法测定硫酸基的含量、糖腈乙酸酯衍生物GC法测定单糖的组成、分子排阻HPLC法测定纯度及分子量,采用IR法、高碘酸氧化、Smith降解分析其结构。结果经分离纯化得到PMF1、PMF2、PMF3等多糖组分;PMF1由木糖、半乳糖组成,其摩尔百分比分别为3.26%、96.74%;PMF2仅由葡萄糖组成;PMF3由鼠李糖、岩藻糖、木糖、甘露糖、葡萄糖、半乳糖组成,其摩尔百分比分别为9.31、1.76、13.09、14.18、25.49、36.17。PMF1、PMF2、PMF3的平均分子量分别为71.69、84.33、103.17kDa,均为均一组分,具多糖的红外特征吸收峰,含糖醛酸和硫酸基,存在1→3、1→6或1→2键合吡喃己糖残基,PMF2、PMF3中存在α,β-糖苷键。结论对所纯化的桑葚多糖进行了初步的性质研究与结构分析。关键词:桑葚多糖;分离纯化;理化性质中图分类号:R28文献标志码:A文章编号:1006-0103(2014)04-0401-04DOI:10.13375/j.cnki.wcjps.2014.04.015Separation,purificationandcompositionanalysisofpolysaccharidesfromMorifructusTIANRen-jun(SichuanCollegeofTraditionalChineseMedicine,Mianyang,Sichuan,621000P.R.China)Abstract:OBJECTIVEToexplorethemethodsforseparationandpurificationofpolysaccharidesfromMorifructus(PMF),andinvestigatecompositionandstructureofthepolysaccharides.METHODSThecrudePMFwaspurifiedbyDEAE-52ionexchangecolumnchromatographyandfurtherpurifiedthroughaSephadexG-100columntoaffordthepurifiedpolysaccharide.Thecontentoftotalcarbohydrate,protein,uronicacidandsulfuricradicalweremeasuredbythemethodofsulfuricacid-phenolcoloration,coomass-iebrilliantbluecoloration,sulfate-3-Phenylphenylolassay,gelatin-bariumchlorideassay,respectively.Thecompositionofmono-saccharidewasinvestigatedbyusingthemethodofSugarandacrylicacidesterderivativesofgaschromatography.TherelativemolecularweightwasdeterminedbyHPLC-SEC.ThestructuralcharacterizationsofthepolysaccharideswereconfirmedbyNaIO4oxidation,SmithdegradationandIR.RESULTSThreepolysaccharidesofPMF1,PMF2andPMF3wereobtainedfromcrudepolysaccharides.PMF1wascomposedofxyloseandgalactoseinamolarpercentof3.26and96.74,respectively,andPMF2wasonlycomposedofglucose.PMF3wascomposedofrhamnose,fucose,xylose,mannose,glucoseandgalactosewitha
